This is a complete plywood vanity carcass built from one sheet ripped into 20-1/4"-wide strips, with two enclosed end sections and an open shelf in the middle — sized around the plumbing rather than around it. Cut the center shelf's back edge to clear the supply lines and drain before you build it in, since that cutout has to be sized to your actual plumbing, not a fixed dimension.
Build the plywood carcass first with 3/4" pocket holes, then build the face frame separately — this two-piece approach lets you square the face frame on a flat bench before it ever touches the carcass, which is much easier than framing directly onto an already-assembled box. Attach the face frame to the carcass with finish nails and glue once both are done.
One side gets a drawer, the other a door — build the drawer to fit its actual opening and slides rather than to a fixed dimension, since plywood thickness and face frame reveal both shift the real opening size slightly from the plan. The drawer box is inset 3/4" from the face frame to leave room for the drawer front, and the shaker-style door is built from 1x3 stiles and rails around a 1/2" plywood center panel. Fit the doors and drawer front to your own openings last, after the carcass and face frame are both actually assembled, rather than cutting them to the plan's numbers up front.

Build the basic plywood box with 3/4" pocket holes and 1-1/4" pocket hole screws — sides, bottom, inner dividers, and end shelves.
Cut the center open shelf to clear the plumbing at the back before installing it fixed or on adjustable shelf pins.
Attach the back supports and front-to-back nailers with pocket holes on the back side.
Build the face frame separately with 3/4" pocket holes, then attach it to the carcass with 1-1/4" finish nails and glue.
Attach the drawer slides to the inside of the cabinet with nails and glue, following the hardware's instructions.
Build the drawer box to fit your actual opening and slides using 1/2" pocket holes and 1" pocket hole screws, attaching the plywood bottom with finish nails and glue. Inset the box 3/4" to leave room for the drawer front.
Build the shaker-style door from the stiles and rails around the plywood center panel, sized to fit your actual door opening.
Fit the hinges and hang the door, and attach the drawer front. Add knobs and pulls.
Sand all surfaces and apply primer and paint, or stain and poly.
